چکیده

Lyon, Richard H. (Veterans Administration Hospital, Minneapolis, Minn.), Herman C. Lichstein, and Wendell H. Hall. Effect of Tween 80 on the growth of tubercle bacilli in aerated cultures. J. Bacteriol. 86:280-284. 1963.-The effect of Tween 80 (polyoxyethylene sorbitan monooleate), glucose, and glycerol on aerated and stationary growth of Mycobacterium tuberculosis strain H(37)Ra was examined in Dubos liquid medium. Previous studies established that aeration (rotation) of liquid cultures of M. tuberculosis strains H(37)Ra and H(37)Rv caused an inhibition of growth in a medium containing glucose as a source of carbohydrate. The present studies show that Tween 80 exerts a toxic effect on the growth of tubercle bacilli in aerated cultures when glucose is present in the medium as the sole source of carbohydrate, but not when glycerol is included. The role of hydrolytic products of Tween 80, viz. oleic acid and the polyoxyethylene derivative of sorbitol, is discussed. The hypothesis is submitted that glycerol protects against the growth suppression by aeration because it reduces the concentration of free fatty acids in the medium to subinhibitory levels.

The Effect of Wetting Agents on the Growth of Tubercle Bacilli

Tween 80 and Triton A20 are two water-dispersible, non-ionic, surface-active agents which favor dispersed growth of tubercle bacilli in aqueous media probably by wetting the bacterial surface. Tween 80 is a polyoxyethylene ester of sorbitan monooleate and is liable to enzymatic hydrolysis by lipases. Triton A20 in an arylalkyl polyether of phenol which appears resistant to the known enzymes of ...

Virulence and Morphological Characteristics of Mammalian Tubercle Bacilli

Experimental infection of the mouse can be used for the determination of virulence of cultures of mammalian tubercle bacilli. The relative virulence of such cultures for the mouse is approximately the same as for the guinea pig. Cultures of virulent and avirulent variants of mammalian tubercle bacilli grown in the depth of Tween 80-albumin liquid medium, on the surface of solid agar modificatio...

The Effect of "Tween 80," Bovine Albumin, Glycerol, and Glucose on the Growth of Mycobacterium tuberculosis var. hominis (H37Rv).

In a series of publications (Dubos, 1945; Dubos and Davis, 1946; Davis and Dubos, 1946a; Davis and Dubos, 1946b; Dubos, 1946; Dubos, Davis, Middlebrook, and Pierce, 1946; Dubos, 1947; Davis and Dubos, 1947; Dubos and Middlebrook, 1947) the addition of certain wetting agents to synthetic culture media used for the growth of virulent tubercle bacilli has been advocated. The synthetic nonionic det...

Growth-enhancing factor for human tubercle bacilli using minute inocula.

It has been difficult to cultivate human tubercle bacilli in agar medium unless bovine serum, or a fraction therefrom, was added aseptically, since the factor present in the serum is thermolabile (Dubos and Middlebrook, 1947). On the other hand, egg yolk or whole egg media may be used, but the difficulties in preparing the media render the methods uncertain and cumbersome. Hence, a thermostable...

The Binding of Fatty Acids by Serum Albumin, a Protective Growth Factor in Bacteriological Media

Serum albumin is a protective bacterial growth factor; by binding traces of fatty acid in the media it permits initiation of growth by the smallest possible inocula of tubercle bacilli. Each molecule of albumin binds 3 to 6 molecules of oleic acid (1 to 2 per cent of the weight of the albumin) tightly enough to prevent bacteriostasis, and 9 molecules of oleic acid in equilibrium with a saturate...
